Specificity

Clone 2H15 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects ERK5. It targets an epitope within 18 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the C-terminal region of human ERK5.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell nuclear extract.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected ERK5 in NIH3T3 cell nuclear extract.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected ERK5 in human spleen tissue sections.

Affinity Binding Assay:: A representative lot of this antibody bound ERK5 peptide with a KD of 1.3 x 10-8 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected recombinant Human ERK5.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected GST-tagged recombinant Human ERK5.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected ERK5 in one million A549 and HeLa cells.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected ERK5 in Human spleen tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Mitogen-activated protein kinase 7 (UniProt: Q13164; also known as EC:2.7.11.24, MAP kinase 7, MAPK 7, Big MAP kinase 1, BMK-1, Extracellular signal-regulated kinase 5, ERK-5) is encoded by the MAPK7 (also known as BMK1, ERK5, PRKM7) gene (Gene ID: 5598) in human. ERK5 is an unusual protein kinase with a kinase domain (aa 55-347) and a transcriptional transactivation domain (aa 664-789). It is expressed in many adult tissues and higher expression is observed in heart, placenta, lung, kidney, and skeletal muscle. Although its kinase domain  displays high sequence identity with ERK1/2, they tend to have distinct substrates and unlike ERK1/2, it has a large C-terminal extension that contains a NuCLEAR localization signal (aa 505-539) and the transcriptional transactivation domain. ERK5 plays a role in various cellular processes, including cell proliferation, differentiation, and cell survival. Its upstream activator is MAP2K5 and following activation, it translocates to the nucleus and phosphorylates various downstream targets. It is activated by Dual® phosphorylation of threonine 219 and tyrosine 221. ERK5 is also important for endothelial function and maintenance of blood vessel integrity. ERK5-deficient mice die around embryonic day 10.5 because of cardiovascular defects and angiogenic failure in embryonic and extraembryonic tissues.  Four isoforms of ERK5 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
