General description

Linearity
The method is linear in the range from 0.02 mg nitrite or nitrate/l sample solution to 5.00 mg nitrite or nitrate/l sample solution.

Detection limit: 0.02mg/l for nitrite and nitrate

Specificity

Under the provided conditions, nitrate reductase reacts specifically with nitrate ions.

Application

Semimicro method for the determination of nitrite and/or nitrate to investigate water, waste water, environmental samples, plant material, foodstuffs, drugs, cosmetics, and biological samples. Nitrite/Nitrate, Colorimetric Test was used in a study done to estimate the in vivo effects of myrtle oil (Myrtii oleum) on the antioxidant enzymes such as catalase and superoxide dismutase in alloxan and normoglycaemic diabetic rabbits.

Preparation Note

Working solution: Preparation of the solutions
  • Use the contents of bottle #1 undiluted, bring to 15 to 25 °C before use.
  • Place one Coenzyme-tablet in a beaker and dissolve in 3 ml of the solution from bottle 1. Remove the Coenzyme-tablet using the tweezers provided. The resulting solution is reaction mixture 2 and is sufficient for 12 nitrate determinations, bring to 15 to 25 °C before use.
  • Dissolve the contents of one bottle #3 in 0.7 ml of double-dist. water to give solution 3.
  • Use the contents of one bottle #4 undiluted.
  • Use the contents of one bottle #5 undiluted.

Storage conditions (working solution):
  • Prepare Reaction Mixture 2 immediately before use.
  • Solution of Nitrate reductase is stable at 2 to 8 °C for 2 weeks.
  • Solution of Color reagent I is stable at 2 to 8 °C for 3 months.
  • Solution of Color reagent II is stable at 2 to 8 °C for 3 months.
