General description

Inhibitor of nuclear factor kappa-B kinase subunit epsilon (UniProt: Q14164; also known as EC: 2.7.11.10, I-kappa-B kinase epsilon, IKK-E, IKK-epsilon, IkBKE, Inducible I kappa-B kinase, IKK-i) is encoded by the IKBKE (also known as IKKE, IKKI, KIAA0151) gene (Gene ID: 9641) in human. IKK-epsilon is a homodimeric serine/threonine kinase that is highly expressed in spleen, thymus, peripheral blood leukocytes, pancreas, and placenta. Its activity is induced by lipopolysaccharide and TNF-alpha. It plays an essential role in regulating inflammatory responses to viral infection, through the activation of the type I IFN, NF-kappa-B, and STAT signaling. IKK-epsilon can be either be auto-phosphorylated or phosphorylated by IKBKB/IKKB and phosphorylation at serine172 is enhanced by the interaction with ATP-dependent RNA helicase DDX3X. Its ATP-binding site resides in amino acids 15 to 23. IKK-epsilon is reported to phosphorylate inhibitors of NF-kappa-B leading to their ubiquitination and degradation. Following activation of viral RNA sensors, such as RIG-I-like receptors, it associates with DDX3X and phosphorylates interferon regulatory factors (IRFs), IRF3 and IRF7, as well as DDX3X. This activity allows subsequent homodimerization and nuclear translocation of the IRF3 leading to transcriptional activation of pro-inflammatory and antiviral genes including IFNB. In order to establish such an antiviral state, IKBKE forms several different complexes whose composition depends on the type of cell and cellular stimuli. Two isoforms of IKK-epsilon have been described that are produced by alternative splicing.
