Specificity

Clone 1O12 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Claudin-1. It targets an epitope within 24 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal region of human Claudin-1.

Application

Quality Control Testing

Evaluated by Western Blotting in MDCK cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Claudin-1 in MDCK cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Claudin-1 in A431, A549 cell lysates and 1:10,000 dilution detected Claudin-1 in RT4 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound Claudin-1 with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Claudin-1 in human skin tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Claudin-1 in A431 cells.

Target description

Claudin-1 (UniProt: O95832; also known as Senescence-associated epithelial membrane protein) is encoded by the CLDN1 (also known as CLD1, SEMP1, UNQ481/PRO944) gene (Gene ID: 9076) in human. Claudins function as major constituents of the tight junction complexes that regulate the permeability of epithelia. While some claudin family members play essential roles in the formation of impermeable barriers, others mediate the permeability to ions and small molecules. Often, several claudin family members are co-expressed and interact with each other, and this determines the overall permeability. Claudin-1 is a multi-pass membrane protein with three cytoplasmic domains, four transmembrane domains, and two extracellular domains. It is strongly expressed in liver and kidney and its expression is also observed in heart, brain, spleen, lung, and testis. It can form homo- and heteropolymers with other claudin family members. It also directly interacts with TJP1/ZO-1, TJP2/ZO-2 and TJP3/ZO-3 and residues 210 and 211 are required for this interaction. Claudin-1 is shown to be essential for preventing the paracellular diffusion of small molecules through tight junctions in the epidermis and is required for the normal barrier function of the skin. It is required for normal water homeostasis and prevents excessive water loss through the skin. In human liver cells, claudin-1 is reported to promote invasive behavior by activating the c-Abl-PKC signaling pathway. CLDN1 knock-out mice die at the first day of birth due to increased trans-epidermal water loss and human subjects lacking claudin-1 suffer from the Neonatal Ichthyosis-Sclerosing Cholangitis (NISCH) syndrome that includes an ichthyosiform skin phenotype. Claudin-1 is also known to associate with CD81 and the claudin-1-CD81 receptor complex is reported to be essential for hepatitis C virus (HCV) entry into host cell. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
