Specificity
Clone 1D11 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Low-density lipoprotein receptor-related protein 6 (LRP6). It targets an epitope within 23 amino acids from the C-terminal, cytoplasmic domain.
Immunogen
KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal, cytoplasmic region of human Low-density lipoprotein receptor-related protein 6 (LRP6).
Application
Quality Control Testing
Evaluated by Western Blotting with FLAG®-tagged recombinant Human LRP6 protein.
Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected FLAG®-tagged recombinant Human LRP6 protein.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LRP6 in human kidney tissue sections.
Enzyme Immunoassay Analysis: Various dilutions of this antibody detected recombinant human LRP6.

Target description
Low-density lipoprotein receptor-related protein 6 (UniProt: O75581; also known as LRP-6) is encoded by the LRP6 gene (Gene ID: 4040) in human. LRP-6 is a disulfide-linked homodimeric, single-pass type I transmembrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 20-1370), a transmembrane domain (aa 1371-1393), and a cytoplasmic tail (aa 1394-1613). It is a component of the Wnt-Fzd-LRP5-LRP6 complex that triggers β-catenin signaling. LRP-6 is widely co-expressed with LRP5 during embryogenesis and in adult tissues. Its levels decrease upon Wnt3A stimulation. LRP-6 contains five PPPSP motifs that play a central role in signal transduction following phosphorylation and activation of Wnt signaling pathway. Dual phosphorylation of cytoplasmic PPPSP motifs by GSK3 and CK1 is shown to be essential for AXIN1-binding, and subsequent stabilization and activation of β-catenin via preventing GSK3-mediated phosphorylation of β-catenin. Upon Wnt signaling, LRP-6 undergoes a cycle of caveolin- or clathrin-mediated endocytosis and plasma membrane location and it undergoes phosphorylation, aggregation, and co-localization with AXIN1 and GSK-3β at the plasma membrane in LRP-6 signalosomes. It is released from the endoplasmic reticulum following palmitoylation. However, in the absence of palmitoylation mono-ubiquitination retains it in the endoplasmic reticulum. LRP-6 is phosphorylated on serine 1490 by CDK14 during G2/M phase leads to regulation of the Wnt signaling pathway during the cell cycle. It is phosphorylated on threonine 1479 by casein kinase I (CK1, which is y TMEM198. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
