General description

A pyrazolopyrimidine compound that blocks ADP release from the actomyosin complex and acts as a potent, reversible and uncompetitive inhibitor of actin-stimulated ATPase activity of myosin V (Ki = 6.3 and 5.5 µM by single- and double-headed myosin V). Displays excellent selectivity over myosin II and myosin IV (<10% inhibition at 50 µM) and only minimally inhibits the kinase activity of Cdk1/B, CHK1, Plk1, Abl, ERK2, CKII and Aurora-B (<10% inhibition at 100 µM).

Neurons have a membrane periodic skeleton (MPS) composed of actin rings interconnected by spectrin. Here, combining chemical and genetic gain- and loss-of-function assays, we show that in rat hippocampal neurons the MPS is an actomyosin network that controls axonal expansion
