Specificity

Recognizes metabotropic glutamate receptor 1a (mGluR1a). On western blots of rat brain microsomes, the antibody labels a single band migrating at 140 kDa, corresponding to mGluR1alpha (Testa, 1998). The antibody recognizes a 140 kDa band in extracts from transfected cells expressing mGluR1alpha but not in extracts from transfected cells expressing mGluR5 (Marino, 2001).

Western blot: 0.25-1.0 μg/mL. Detects a band migrating at approximately 140 kDa. The use of membrane preparations and protease inhibitors is highly recommended.

Immunohistochemistry: AB1551 can be used for immunocytochemistry using paraformaldehyde or paraformaldehyde/glutaraldehyde fixed tissue with light and electron microscopy. Cryostat and vibratome sections can be used with or without Triton X-100 treatment. Final concentrations recommended are 0.5-2.0 μg/mL.

The antibody has not been tested for use in immunoprecipitation. However, antibodies to the carboxy termini of other glutamate receptors are very effective in immunoprecipitation.

Optimal working dilutions must be determined by end user.

Storage and Stability

Maintain lyophilized material at 2–8°C or -20°C. Reconstitute to 500 μL with sterile distilled water. Store reconstituted material frozen (-20°C or -80°C) in undiluted aliquots for up to six months. Avoid repeated thawing and refreezing. Diluted antibody containing 0.1% sodium azide for Western blot analysis can be stored for several weeks and reused repeatedly.
