Cell Line Origin

Rat medullary thyroid carcinoma

Cell Line Description

Derived from the non-clonal line rMTC 6-23 originally established from a transplantable WAG/RIJ rat medullary thyroid carcinoma. The cell line synthesises and secretes both neurotensin and calcitonin. Its clonal derivation demonstrates that a population derived from a single type of C-cell has the capability of producing both peptides. Secretion of these peptides has been shown to be stimulated by calcium, KC1, glucagon and catecholamines.

Culture Medium

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS). Cells need to be grown on flasks coated with matrigel or any other attachment factor

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. For cell attachment culture vessels need to be coated with collagen type IV e.g. Sigma product C-5533. Soak in a sol
