General description

GHRH/GRH (growth hormone releasing hormone) is an endocrine peptide hormone composed of 44 amino acids† It exists in two forms in the hypothalamus, namely GRH(1-44)-NH2 and GRH(1-40)-OH. However, the full biological functionality of this hormone resides in the first 29 amino acids itself. GRH(1-29)-NH2 is a C-terminal shortened peptide produced from GHRH.

Biochem/physiol Actions

Amidated GRF fragment that is equipotent to GRF in releasing somatotropin from anterior pituitary.
GHRH/GRH (growth hormone releasing hormone) is released from the hypothalamus from where it reaches the anterior pituitary and binds to its receptor, thus, eventually leading to the release of GH (growth hormone) into the plasma.

The Journal of clinical investigation, 83(5), 1533-1540 (1989-05-01)
The plasma enzyme responsible for primary proteolytic cleavage of growth hormone-releasing hormone (GRH) at the 2-3 amino acid bond was characterized. Native GRH[GRH(1-44)-NH2 and GRH(1-40)-OH], and COOH-terminally shortened fragments [GRH(1-32)-NH2 and GRH(1-29)-NH2] were rapidly cleaved, while GRH(2-32)-NH2 was not degraded

Benign prostatic hyperplasia often affects aging men. Antagonists of the neuropeptide growth hormone-releasing hormone reduced prostate weight in an androgen induced benign prostatic hyperplasia model in rats. Luteinizing hormone-releasing hormone antagonists also produce marked, protracted improvement in lower urinary tract

Treatment of colon cancer with an antagonist of growth hormone-releasing hormone (GHRH), JMR-132, results in a cell cycle arrest in S-phase of the tumor cells. Thus, we investigated the effect of JMR-132 in combination with S-phase-specific cytotoxic agents, 5-FU, irinotecan
