General description

215 amino acids; comprises two homologous HMG boxes, rich in basic amino acids, and an acidic tail at the carboxy-terminus
HMG-1 (high mobility group protein) is a nuclear protein of molecular weight 30kDa. It is secreted from macrophages, monocytes and necrotic cells. It is ubiquitously expressed. The protein has basic HMG (high mobility group) box domains and an acidic carboxyl-terminus.

Application

HMG-1 (high-mobility-group protein) human has been used:
  • to prepare a standard curve for measuring HMG1 using ELISA (enzyme-linked immunosorbent assay) test
  • to study the apoptotic ratios of Lewis cells in presence of exogenous HMG-1
  • to study the changes in Th (T helper)-17 cell expansion in the presence of HMG-1 under in vitro conditions
  • for the migration assay of GM7373 cells

Biochem/physiol Actions

HMG-1 (high mobility group protein) is a non-histone chromosomal protein, which promotes the sequence specific DNA attachment of proteins. HMG1 binds DNA in a non-sequence specific manner. It is involved in DNA replication, transcription and repair. In addition, it is up-regulated in cancers and is associated with cellular growth, vasculogenesis and apoptosis regulation. In the presence of infection or injury, HMG1 is secreted and mediates inflammation. It is associated with the pathogenesis of autoimmune diseases, such as rheumatoid arthritis (RA), inflammatory bowel disease (IBD) and systemic lupus erythematosus (SLE).

Upregulated high-mobility group box 1 (HMGB1) has been found in many diseases. Nevertheless, the function of HMGB1 on modulating the proliferation of lung cancer cells (Lewis cells) and inhibiting apoptosis is poorly understood, as well as the involved intracellular signalling.
