General description

Trypan Blue (TB), an anionic hydrophilic azo dye, crosses only the cell membranes of dead cells, thereby staining dead tissues/cells blue. It is used in quantitative microscopy for a dye exclusion staining technique to differentiate between live and dead mammalian cells.
Trypan blue is a dye used to distinguish between live and dead cells. It is a vital stain that is not absorbed by healthy viable cells, but stains cells with a damaged cell membrane. This way, only dead cells can be counted. The method is sometimes referred to as the dye exclusion method.

Application

Trypan Blue solution has been used:
  • in cell viability assay to count viable and dead cells
  • in rescue assay to count viable cells
  • in trypan blue exclusion counting method to determine proliferation curve
  • as the saline control and in the preparation of lipopolysaccharide (LPS) solution to confirm the success of LPS infusion
  • in trypan blue dye exclusion test to determine gastric epithelial cells (GEC) viability
  • to count GEC cells using a hemocytometer

Preparation Note

Prepared in 0.81% sodium chloride and 0.06% potassium phosphate, dibasic.
