Specificity

Clone 3F11 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Doublecortin. It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Doublecortin.

Application

Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Doublecortin in SH-SY5Y cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Doublecortin in mouse brain tissue sections.

Immunocytochemistry Analysis: A 1:10 dilution from a representative lot detected Doublecortin in Neuro-2a and U-87 MG cells.

Immunofluorescence Analysis: A 1:10 dilution from a representative lot detected Doublecortin in frozen mouse brain tissue sections.

Anti-Doublecortin, clone 3F11 ZooMAb®, Cat. No. ZRB1863, is a recombinant Rabbit monoclonal antibody that detects Doublecortin and tested for use in Western Blotting, Immunofluorescence, Immunocytochemistry, and Immunohistochemistry (Paraffin).

Target description

Neuronal migration protein doublecortin (UniProt: O43602; also known as Doublin,Lissencephalin-X, Lis-X) is encoded by the DCX (also known as DBCN, LISX) gene (Gene ID: 1641) in human. Doublecortin is a microtubule-associated protein that is required for initial steps of neuronal dispersion and cortex lamination during cerebral cortex development. It is expressed almost exclusively in immature neurons. Hence, it has emerged as an important marker of neurogenesis. It is highly expressed in newly produced cells in the neurogenic zones; the sub ventricular zone along the lateral ventricle and the subgranular zone (SGZ) of the dentate gyrus. Doublecortin binds to microtubules in migrating cells and as a result promote the movement of these cells. It is reported to direct neuronal migration by regulating the organization and stability of microtubules. Doublecortin may act by competing with the putative neuronal protein kinase DCLK1 in binding to a target protein and may participate in a signaling pathway that is crucial for neuronal interaction before and during migration. It undergoes phosphorylation by MARK1, MARK2, and PKA and the phosphorylation regulates its ability to bind microtubules. Doublecortin is reported to be involved in cancer metastasis where unstable interactions between doublecortin and microtubules destabilize cytoskeletal organization leading to disorganized movements of cells that can lead to the uncontrolled migration of cancer cells. Mutations in DCX gene are reported to cause abnormal migration of neurons during development and disrupt the layering of the cortex, leading to epilepsy, cognitive disability, subcortical band heterotopia. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
