Specificity

Clone 1G2 is a Rabbit recombinant monoclonal antibody that specifically detects Cathepsin L. It targets an epitope within 14 amino acids from the C-terminal half. It detects procathepsin L, mature Cathepsin L, and Cathepsin L heavy chain.

Immunogen

KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal half of human Cathepsin L.

Application

Quality Control Testing

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Cathepsin L1 in PC12 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Cathepsin L1 in A549 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound Cathepsin L1 peptide with a KD of 4.1 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Cathepsin L1 in A549 cells

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Cathepsin L1 in human kidney tissue sections.

Target description

Procathepsin L (UniProt: P07711; also known as EC:3.4.22.15, Cathepsin L, Major excreted protein, MEP) is encoded by the CTSL (also known as CTSL1) gene (Gene ID: 1514) in human. Cathepsin L is a lysosomal protease that is synthesized as a procathepsin with a signal peptide (aa 1-17) and two propeptides (aa 18-113 activation peptide; and 289-291) that are cleaved off to produce mature active enzyme. After the removal of these peptides, cathepsin can exist in three forms: cathepsin L (aa 114-333), cathepsin L heavy chain (aa 114-288) and cathepsin L light chain (aa 292-333). The heavy and light chains are connected by disulfide bonds. Active cathepsin L is a thiol protease with endopeptidase activity and preferentially cleaves peptide bonds with aromatic residues in P2 position and hydrophobic residues in P3 position. It is catalytically active in the pH range 3 to 6.5. It participates in overall degradation of proteins in lysosomes and plays a critical for normal cellular functions such as general protein turnover, antigen processing and bone remodeling. It is also Involved in the solubilization of cross-linked thyroglobulin and in subsequent release of thyroxine (T4) by limited proteolysis of thyroglobulin in the thyroid follicle lumen. Cathepsin L has been implicated in several pathological conditions, including invasion and tumor metastasis, atherosclerosis, inflammation, and degradation of extracellular matrix that is reported to be an important process in entry of SARS-CoV-2 into host cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
