General description

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

Immunogen

KLH-conjugated linear peptide corresponding to a region near the N-terminus of human dimethyl Histone H3 methylated at Lys36.

Application

Anti-dimethyl Histone H3 Antibody (Lys36) is a highly specific rabbit polyclonal antibody, that targets Histone H3 & has been tested in western blotting, Dot Blot & Chromatin IP (ChIP).
Dot Blot Analysis: A 1:10,000 dilution from a representative lot detected dimethyl Histone H3 (Lys36), but not unmethylated Histone H3 or other peptides corresponding to modified histones.

Chromatin Immunoprecipitation (ChIP) Analysis: A representative lot imunoprecipitated dimethyl Histone H3 (Lys36) in HeLa cell lysate.

Quality

Evaluated by Western Blotting in HeLa acid extract.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected dimethyl Histone H3 (Lys36) in 10 µg HeLa acid extract and demonstrated a loss of signal in 0.5 µg of unmethylated recombinant Histone H3.
