General description

The gene SFRP1 (secreted frizzled related protein 1) is mapped to human chromosome 8p22–p12, a region frequently deleted in breast cancer. It is highly expressed in normal breast tissue.

Biochem/physiol Actions

Secreted Frizzled-related protein 1 (SFRP1) is a secreted inhibitor of Wnt signaling pathway. DNA damage or oxidative stress causes cellular senescence, which leads to overexpression of sfrp1. This protein induces senescence phenotypes via the inactivation of Wnt signaling and activation of the retinoblastoma (Rb) pathway. Mutations or aberrant methylation in this gene impair the induction of senescence in several cancers, including breast cancer, bladder cancer, and colorectal tumorigenesis.

Physical form

Lyophilized from 10 mM Sodium Phosphate pH 7.0.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
