Specificity

Clone RMT3-23 is a ZooMAb® rat recombinant monoclonal antibody that specifically detects TIM-3.

Immunogen

Recombinant fragment corresponding to the first 191 amino acids from of Mouse TIM-3 conjugated to the Fc portion of mouse IgG2a.

Application

Quality Control Testing

Evaluated by Western Blotting in Rat thymus tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TIM-3 in Rat thymus tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TIM-3 in Human thymus tissue lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TIM-3 in L5178Y cells transfected with TIM-3.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected TIM-3 in one million L5178Y cells transfected with TIM-3.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Hepatitis A virus cellular receptor 2 homolog (UniProt: Q8VIM0; also known as HAVcr-2, T-cell immunoglobulin and mucin domain-containing protein 3, TIMD-3, T-cell immunoglobulin mucin receptor 3, TIM-3, T-cell membrane protein 3, CD366) is encoded by the Havcr2 (also known as Tim3, Timd3) gene (Gene ID: 171285) in murine species. TIM-3 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 20-193), a transmembrane domain (aa 194-214), and a cytoplasmic domain (aa 215-281). Its expression is observed in T-helper type 1 cells, but not in naïve T cells. However, its expression is up-regulated as they differentiate into T helper type 1 cells. It is also expressed by differentiated type 1 CD8+ cytotoxic T-cells, peritoneal exudate macrophages, monocytes, and splenic dendritic cells. It serves as a receptor for galectin-9 (LGALS9), phosphatidylserine, HMGB1, and CEACAM1. Binding of LGALS9 is proposed to be involved in innate immune response to intracellular pathogens. Phosphorylation of TIM-3 on tyrosine 256 is reported to increase by binding to LGALS9. Phosphorylation on tyrosine 256 and 263 also couples it to TCR signaling pathways. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rat monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

30 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
