General description

The NTRK2 (neurotrophic tyrosine kinase receptor type 2) gene is mapped to human chromosome 9q21.33 and is predominantly expressed in subcutaneous fat and also expressed in visceral fat. Insulin receptor subfamily. NTRK2 / TrkB contains two Ig-like C2-type (immunoglobulin-like) domains, two LRR (leucine-rich) repeats, one LRRCT domain, one LRRNT domain, one protein kinase domain. NTRK2 / Trk-B is expressed in the central and peripheral nervous system. The catalytic activity of NTRK2 is “ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate”.

Biochem/physiol Actions

Neurotrophic tyrosine kinase receptor type 2 (NTRK2) is an oncogenic receptor tyrosine kinase. Abnormal expression of the NTRK2 gene is associated with several cancers including colorectal, breast and papillary thyroid cancer, lung adenocarcinoma and oral squamous cell carcinoma. NTRK2 also known as tropomyosin-related kinase B (TRKB) and is activated by binding of a number of neurotrophins including brain-derived neurotrophic factor(BDNF), neurotrophin 3 (NT-3) and neurotrophic ⅘ (NT-4/5). NTRK2 is involved in the development and the maturation of the central and the peripheral nervous systems through regulation of neuron survival, proliferation, migration, differentiation, and synapse formation and plasticity.

Physical form

Lyophilized from 0.22 μm filtered solution in 50 mM tris, 100 mM glycine, pH 7.0. Normally Mannitol or Trehalose is added as protectants before lyophilization.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 μg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
