Application

C8 Dihydroceramide (d18:0/8:0) has been used as an internal standard to quantify the lipid extracted from human plasma by liquid chromatography mass spectrometry during high-throughput plasma lipidomics.[1]

Biochem/physiol Actions

C8 Dihydroceramide (d18:0/8:0) or N-octanoyl-D-erythro-sphinganine, is a synthetic dihydroceramide. Dihydroceramide acts as a precursor for de-novo ceramide synthesis.[2] C8 Dihydroceramide mimics γ-tocotrienol (γTE) in stimulating NF-κB-target protein A20 expression, increasing endoplasmic reticulum (ER) stress and reducing tumor necrosis factor (TNF)-triggered nuclear factor kappa B (NF-κB) activation.[3]

Vitamin E gamma-tocotrienol inhibits cytokine-stimulated NF-kappaB activation by induction of anti-inflammatory A20 via stress adaptive response due to modulation of sphingolipids

Gaucher disease (GD) is an inherited metabolic disorder characterised by impaired catabolism of the glycosphingolipid, glucosylceramide. The deacetylated derivative, glucosylsphingosine (GluSph, lyso-Gb1) has materialised as a biomarker for GD. Further appraisal of the clinical utility of GluSph is required in

NF-κB plays a central role in pathogenesis of inflammation and cancer. Many phytochemicals, including γ-tocotrienol (γTE), a natural form of vitamin E, have been shown to inhibit NF-κB activation, but the underlying mechanism has not been identified. In this study
Low serum levels of (dihydro-) ceramides reflect liver graft dysfunction in a real-world cohort of patients post liver transplantation

Patients after orthopic liver transplantation (OLT) are at risk of developing graft dysfunction. Sphingolipids (SL's) have been identified to play a pivotal role in the regulation of hepatocellular apoptosis, inflammation and immunity. We aimed to investigate the serum SL profile
