General description

Protein phosphatase 1K (PPM1K) is a mitochondrial enzyme which is activated by Mn2+ ions. It is part of the branched-chain α-ketoacid dehydrogenase complex (BCKDC) and the gene encoding this protein is localized on human chromosome 4q22.

Application

Biochem/physiol Actions

Protein phosphatase 1K (PPM1K) functions in the metabolism of branched-chain amino acids (BCAAs). It is involved in the oxidative decarboxylation of branched-chain α-ketoacids from isoleucine, leucine and valine. PPM1K binds to 24 lipoyl-bearing dihydrolipoyl transacylase (E2b) which forms the core of the branched-chain α-ketoacid dehydrogenase complex (BCKDC). Its activity is stimulated by this binding which is between the carboxyl-terminal linker of E2b and PPM1K.
