The separation and characterization of biomacromolecules presents a distinct analytical challenge due to the intricate nature of the analytes. This complexity arises from the numerous modifications to these analytes in each sample, including phosphorylation, methylation, and glycosylation, among others. The heterogeneity of individual analytes, combined with a biomolecule's tendency to interact with silica-based stationary phases, can lead to significant broadening and tailing of chromatographic peaks. Additionally, conventional electrospray ionization mass spectrometers may not effectively ionize large proteins. Furthermore, poorly separated protein mixtures can result in further ion suppression. Consequently, the demand for highly efficient, robust, and reproducible analytical methods is not merely a suggestion but a necessity for the accurate characterization of proteinaceous samples.

This seminar will concentrate on strategies and techniques to enhance efficiency and resolution when employing various chromatography modes to analyze biomacromolecules. It will delve into method development, troubleshooting aspects, and chromatographic considerations for biomacromolecule characterization suitable for mass spectrometry (MS) and Multi-Angle Light Scattering (MALS) detection platforms.  By the end of the seminar, participants will gain insight into:

  • Selecting and understanding the importance of phase chemistry and particle morphology in biomolecule analysis
  • Best practices for selecting appropriate mobile phase systems for each chromatography mode
  • Diagnosing the root causes of issues encountered when troubleshooting biomolecule separations

Egidijus Machtejevas was born in Kaunas, Lithuania where he studied chemistry, biotechnology and separation sciences (1991-2001) at Kaunas University of Technology, Lithuania. Also, as a research student spent a year in Prof. Stelan Hjerten laboratory in Uppsala University (Sweden). After gaining his PhD in analytical chemistry 2001-2007 he worked as a post-doc with Prof. Klaus Unger at Mainz University, Germany. Also two years spent at AstraZeneca (Sweden) where he set up and validated several various multidimensional systems for peptidomic analysis of different biological fluids. He joined the R&D Department at Merck KGaA, Darmstadt, Germany in 2008, in 2009 took a marketing manager position for North America, 2010 - 2018 product manager for chromatography. Currently, he is a Senior Technical Advisor in the Analytical Chromatography Workflows group. Egidijus Machtejevas has twenty-five scientific papers and thirteen book chapters to his name, and his major focus areas include chromatography, multidimensional liquid chromatography, proteomics and mass spectrometry.

Corey Meadows obtained his PhD in physical chemistry at University of California, Berkeley in 2014.  He served as an analytical development scientist in the renewables and biopharmaceutical industries, with a focus on HPLC, LC-MS, and GC-MS techniques for small and large molecule analyses.  While working within the biopharmaceutical industry, he focused primarily on antibody and protein therapeutic characterization using high resolution UHPLC-MS methods.  His experience in the renewables industry focused on implementing chromatography and mass spectrometry-based techniques for small molecule metabolite quantitation.  He represents Tosoh Biosciences as a Regional Product Specialist, focusing on application support and product development for analytical columns and multi-angle light scattering (MALS) detectors.
