General description

The Tribbles family of proteins are kinase-like proteins that contain a kinase-like domain, but lack either a catalytic domain or a protein-protein interaction domain (2). They are believed to regulate signal processing for various pathways including MAPK, Akt, and NFkB that are involved in various aspects of development, diabetes, and cancer (1,2,3). Their involvement has been reported to regulate a number of pathways that include mitosis, cell activation, and apoptosis. They are are a family made up of three family members. Tribbles-1 (TRIB1, SKIP1, G-protein-coupled receptor induced protein, and phosphoprotein regulated by mitogenic pathways) is a modulator of MAPK action. It has been shown to directly interact with the upstream kinases MEK1 and MKK4 of Erk and JNK, respectively(1). Tribble-2 (Trib2, TRB2) has been shown to interact with Akt(2). Tribbles-3 (TRIB3, TRB3. NIPK, SKIP3) has been shown to interact with Akt and the transcription factors ATF4 and NFkB p65 (RelA). It is interaction with RelA is thought to be negative regulation. Interestingly, it has been reported to regulate Akt activation via insulin(3).
