General description

An internal standard for clinical testing of Cyclosporin A by LC-MS/MS. This Certified Spiking Solution® is suitable for critical quantitative applications in clinical and diagnostic testing such as therapeutic drug monitoring assays to ensure patients remain within Cyclosporin A′s narrow therapeutic range. The Snap-N-Spike® format allows laboratories to eliminate weighing operations of these hazardous substances, significantly reducing occupational exposure hazards related to handling of acutely toxic powders, and provides the ability to spike into their matrix of choice just before use.

Application

  • High-Performance Liquid Chromatography for Cyclosporin D: A study developed a reversed-phase high-performance liquid chromatography method for the determination of cyclosporin A in human and mouse plasma, using Cyclosporin D as internal standard, highlighting its applications in drug discovery and bioavailability studies (Bardelmeijer et al., 2001).

  • Mass Spectrometric Analysis of Cyclosporins: Research focused on the thermospray high-performance liquid chromatography/mass spectrometric determination of cyclosporins, including Cyclosporin D, contributing to advancements in the analysis of immunosuppressant drugs and their pharmacological profiling (Abián et al., 1992).

Using solid-phase extraction columns and "high-performance" liquid-chromatographic (HPLC) analysis, we could determine cyclosporin A and nine of its metabolites in blood, bile, and urine. To facilitate calculations of concentrations of cyclosporin A and its metabolites from the chromatograms, we used

The cyclosporines are a family of cyclic endecapeptides that cause a profound suppression of primary immune stimulation both in vitro and in vivo. Recently, the regulatory protein calmodulin (CaM) has been implicated as a target for cyclosporin A (CsA) binding.
