General description

60S ribosomal protein L5 (UniProt P46777; also known as RPL5) is encoded by the RPL5 (also known as DBA6, MSTP030) gene (Gene ID 6125) in human. Ribosomal proteins (RPs) are normally involved in the assembly of the translational machinery-ribosomes essential for protein production. RPL5, RPL11 and the 5S rRNA comprise the 5S ribonucleoprotein particle (RNP), an essential subcomplex of the large ribosomal subunit. RPL5 binds the 5S rRNA, the 5S rRNA/RPL5 complex then localizes to the nucleolus, where it binds RPL11 and is integrated into the ribosome. In response to ribosomal stress (RS) or nucleolar stress, the L5/L11/5S pre-ribosomal complex is redirected to inhibit MDM2 E3 ligase activity, thereby promoting p53 stabilization and p53-dependent cell cycle arrest. RPL5 and RPL11 can also bind TAp73 N-terminal TA domain in an RS-independent manner and thereby prevent MDM2 association with TAp73 target gene promoters, resulting in enhanced TAp73 transcriptional activity and TAp73-dependent apoptosis induction and cell cycle arrest.

Specificity

This rabbit polyclonal antibody targets a highly conserved sequence within the C-terminal half of 60S RPL5 from multiple species.

Immunogen

KLH-conjugated linear peptide corresponding to an internal sequence within the C-terminal half of human 60S RPL5.

Application

Anti-60S RPL5, Cat. No. ABE1439, is a highly specific rabbit polyclonal antibody that targets 60S ribosomal protein L5 and has been tested in Immunocytochemistry and Western Blotting.
Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected 60S RPL5 in human (A431, HeLa, HepG2) and mouse (NIH/3T3) cells.

Western Blotting Analysis: 1 µg/mL from a representative lot detected 60S RPL5 in 10 µg of human (A431 epidermoid carcinoma & HUVEC) and rat (L6 myoblast) cell lysates.
