Target description

Integrin alpha-L (UniProt: P20701; also known as CD11 antigen-like family member A, Leukocyte adhesion glycoprotein LFA-1 alpha chain, LFA-1A, Leukocyte function-associated molecule 1 alpha chain, CD11a) is encoded by the ITGAL (also known as CD11A) gene (Gene ID: 3683) in human. Integrins are heterodimeric, integral membrane proteins composed of a variable -subunit and a conserved β-subunit that function in cell surface adhesion and signaling. They contain a large extracellular domain responsible for ligand binding, a single transmembrane domain, and a cytoplasmic domain. The exact combination of various and β-subunits dictates the binding specificity of integrins to different ECM components. Although both subunits are required for adhesion, the binding specificity primarily depends on the extracellular portion of the -subunit. CD11a is a single-pass type I membrane glycoprotein that noncovalently associates with β2 integrin to forms a heterodimer and serve as a receptor for ICAM1, ICAM2, ICAM3 and ICAM4. CD11a is considered as a pan-leukocyte marker and is expressed on B and T lymphocytes, monocytes, macrophages, neutrophils, basophils, and eosinophils. It is involved in generation of common lymphoid progenitor cells in bone marrow. In resting T-cells, about 40% of CD11a is constitutively phosphorylated and phosphorylation causes conformational changes needed for ligand binding and is considered to be essential for activation by some physiological agents. CD11a is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off in the mature form. It has a large extracellular domain (aa 26-1090), a transmembrane domain (aa 1091-1111), and a cytoplasmic domain 1112-1170). Three isoforms of CD11a have been described that are produced by alternative splicing. (Ref.: Bose, TO., et al. (2014). J. Immunol. 193(6); 2863-2872; Fagerholm, SC., et al. (2005). J. Cell Biol. 171(4); 705-715; Sanchez-Madrid, F., et al. (1982). Proc. Natl. Acad. Sci. USA. 79(23); 4489-7493).

Physical form

Purified mouse monoclonal antibody IgG1 in PBS without preservatives.

Reconstitution

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
