Specificity

Target description

Sonic hedgehog protein (UniProt: Q63673; also known as SHH, EC:3.1.-.-, ShhNC, ShhN) is encoded by the Shh (also known as Vhh-1 ) gene (Gene ID: 29499) in rat. The Sonic Hedgehog (Shh) protein is a member of the hedgehog family of signaling molecules which also include Indian Hh and Desert Hh. It is initially synthesized as a precursor protein. The C-terminal domain of the precursor SHH displays an autoproteolysis activity and a cholesterol transferase activity, causing a cleavage of the precursor SHH into an N-terminal fragment (ShhN-product; aa 25-198) as well as the covalent attachment of a cholesterol moiety to the C-terminal end of the N-product. The N-product is the active signaling species, whereas the C-product has no signaling activity. Shh expression is highly regulated and varies depending on the developmental stage and tissue type. In the developing embryo, Shh is expressed in the notochord, floor plate of the neural tube, limb buds, and various other tissues. In adults, Shh expression is more restricted but remains important for tissue homeostasis and repair. The precursor protein is processed in the endoplasmic reticulum and Golgi apparatus before being transported to the cell membrane. The dual-lipidated sonic hedgehog protein N-product (ShhNp) is firmly tethered to the cell membrane where it forms multimers. Dysregulation of Shh signaling can lead to various congenital disorders and diseases. The intercellular signaling triggered by SHH is essential for a variety of patterning events during development including the development of the notochord, neural tube and plate, as well as the polarizing of the anterior-posterior axis of developing limb buds. Because of its importance in patterning during development, defects in SHH are the cause of a number of abnormalities including structural eye and brain abnormalities, teeth and bone malformations and finally limb and bone developmental defects.
