Target description

Insulin gene enhancer protein ISL-1 (UniProt: P61374; also known as Islet-1) is encoded by the Isl1 gene (Gene ID: 64444) in rat. Islet-1 is a DNA-binding transcriptional activator that is expressed in pancreatic islets and is essential for the maturation, proliferation, and survival of the endocrine pancreas. It recognizes and binds to the consensus octamer binding site 5′-ATAATTAA-3′ in promoter of target genes. Its DNA binding region is localized in amino acids 181-240. It also contains two LIM Zinc-binding domains (aa 17-79 and 90-133) and a LIM-binding domain (aa 262-291). It plays a key role in the gene regulatory network essential for retinal ganglion cell (RGC) differentiation. It cooperates with the transcription factor POU4F2 to achieve maximal levels of expression of RGC target genes and RGC fate specification in the developing retina. It is also involved in the specification of motor neurons in cooperation with LHX3 and LDB1. It is reported to be essential for cardiac development and is considered as a marker of one progenitor cell population that give rise to the outflow tract, right ventricle, a subset of left ventricular cells, and a large number of atrial cells. It also controls the expression of fibroblast growth factor and bone morphogenetic protein. Islet-1 expression is a reliable marker for pancreatic endocrine tumors and their metastases.
