Target description

Thyroid peroxidase (Uniprot: P07202; also known as TPO) is encoded by gene TPO (Gene ID: 7173) in human. It is a single-pass type I membrane protein and has an Extracellular domain (19-846 aa), a Transmembrane domain ( 847-871 aa) and a Cytoplasmic domain (872-933 aa). It is primarily found in the thyroid gland and functions in the iodination of tyrosine residues in thyroglobulin and the formation of phenoxy-ester between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine (T3 and T4). TPO is an integral part of the complex process by which the thyroid gland regulates metabolism and other vital bodily functions. Defects in this gene can cause congenital hypothyroidism, congenital goitre and thyroid hormone organification defect IIA. Moreover, TPO is a common target of autoimmune attacks in various thyroid disorders, such as Hashimoto′s thyroiditis and Graves′ disease, which can lead to disruptions in hormone synthesis and subsequent health issues.
