General description

The product is supplied as a ready-to-use one component substrate for peroxidase consisting 2,2′-azino-bis (3-ethylbenz-thiazoline-6-sulfonic acid) in an acidic buffer. The substrate is colorless before reacting with peroxidase and produces a blue-green color reaction product after reacting with it. 1% sodium dodecyl sulfate (SDS) may be used to stop the reaction during end-point assays. Since this substrate produces a soluble reaction product, it is not recommended for immunoblotting or histochemical applications.

Application

2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) has been used as a peroxidase substrate in ELISA experiment. This substrate gives a green color soluble end product. It has also been used as a colloidal preparation spread over the standard media plates, to evaluate the lignin oxidation activity. Presence of green coloration of the substrate exhibits microbial laccase activity.

Biochem/physiol Actions

2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) is a cation and undergoes oxidation in the presence of hydrogen peroxide. The oxidized ABTS is a blue-green product and is measurable colorimetrically. A combination of ABTS, hydrogen peroxide and myoglobin is a standard system employed for measuring total antioxidant capacity(TAC) from samples. ABTS method is flexible at varying pH and most widely used. ABTS is a substrate for phenol oxidase enzyme.
