General description

Progesterone receptor (PGR) belongs to the steroid/thyroid hormone receptor family of ligand-activated transcription factors. It contains many functional domains: the amino-terminal AF1 region, the central DNA-binding domain (DBD) and the carboxyl-terminal ligand-binding domain (LBD).

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Biochem/physiol Actions

Progesterone receptor (PGR) functions as a transcription factor which modulates the transcription of target genes in response to progesterone and other hormones. It is phosphorylated on multiple sites and at times the phosphorylation changes according to the hormone involved. In the absence of hormones, PGR is complexed with heat shock proteins. When the ligand binds, it causes the release of PGR from heat shock proteins. It is then translocated to the nucleus where it binds to its DNA response elements (SREs) and to the components of the transcription machinery. PGR also binds to chromatin templates and results in chromatin remodeling adjacent to the PGR-binding sites.
