Biochem/physiol Actions

Orally active basic fibroblast growth factor (bFGF, FGF2) mimetic with neuroprotective efficacy in vitro and in vivo.
SUN11602 is an orally active basic fibroblast growth factor (bFGF, FGF2) mimetic that protects against glutamate toxicity in primary rat cerebrocortical neuron (0.1-1 μM SUN11602; 150 μM glutamate) and murine hippocampal neuron cultures (0.3-3 μM SUN11602; 100 μM glutamate). When administered in vivo, SUN11602 exhibits therapeutic efficacy in a rat hippocampal-lesion model (1-10 mg/kg p.o. 24h post Aβ1-40 injection) and a murine MPTP-induced nigrostriatal degeneration model (2.5-5 mg/kg/day p.o.).

Small molecules have gained considerable interest in regenerative medicine, as they can effectively modulate cell fates in a spatiotemporal controllable fashion. A continuous challenge in the field represents genuine mimicry or activation of growth factor signaling with small molecules. Here

Basic fibroblast growth factor (FGF-2/bFGF) possesses neuroprotective activity and promotes cell proliferation. In this study, the novel synthetic compound 4-({4-[[(4-amino-2,3,5,6-tetramethylanilino)acetyl](methyl)amino]-1-piperidinyl}methyl)benzamide (SUN11602) exhibited neuroprotective activities similar to those of FGF-2 without promoting cell proliferation. In primary cultures of hippocampal neurons, stimulation
