Biochem/physiol Actions

Cell-permeable, BODIPY-tagged C16 fatty acid analog, a green-fluorescent lipid tracer.

BODIPY (dipyrrometheneboron) fluorophores are non-polar, highly photostable, and exhibit a high fluorescence quantum yield, making them suitable for confocal and two-photon fluorescence microscopy, with excitation and emission at 488 nm and 510 - 520 nm. BODIPY-FL-C16 , a palmitate analog, tagged with BODIPY, is efficiently taken up by Caco-2 cells and aligns with L-FABP and markers for cellular organelles, exhibiting strong binding to liver and intestinal fatty acid-binding proteins (with KD values of 270 and 330 nM, respectively) akin to oleic acid, and its fluorescence intensity markedly rises to 0.87 and 0.72 in the presence of L-FABP and I-FABP, respectively. BODIPY-FL-C16 serves as well to track lipid peroxidation in red blood cell membranes.
