Specificity

Clone 6C19 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Fibrillin-1. It targets an epitope within the N-terminal half.

Immunogen

GST/His-tagged recombinant fragment corresponding to 95 amino acids from the N-terminal half of human Fibrillin-1.

Anti-Fibrillin-1, clone 6C19 ZooMAb, Cat. No. ZRB1019, is a recombinant Rabbit monoclonal antibody that specifically targets Fibrillin-1 and has been tested for use in Affinity Binding Assay, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Fibrillin-1 (UniProt: P35555; also known as FBN1) is encoded by the FBN1 (also known as FBN) gene (Gene ID: 2200) in human. Fibrillin-1 is a structural component of the 10-12 nm diameter microfibrils of the extracellular matrix that conveys both structural and regulatory properties to load-bearing connective tissues. Fibrillin-1-containing microfibrils provide long-term force bearing structural support. Fibrillin-1 is synthesized with a signal peptide (aa 1-24) and a propeptide (aa 25-44) that are cleaved off to generate the mature Fibrillin-1, which can be further cleaved by furin between residues 2731 and 2732 to produce asprosin (aa 2732-2871). Asprosin serves as a hormone that targets the liver to increase plasma glucose levels. It promotes glucose release from hepatocytes by activating protein kinase A. Fibrillin-1 is reported to play a role in tissue homeostasis through specific interactions with growth factors, such as the bone morphogenetic proteins (BMP), growth and differentiation factors, and latent TGF-beta binding proteins. It is also reported to regulate osteoblast maturation by controlling TGF-beta bioavailability and calibrating TGF-beta and BMP levels. It is also shown to negatively regulate osteoclastogenesis by binding and sequestering an osteoclast differentiation and activation factor TNFSF11. Mutations in FBN1 gene are known to cause Marfan syndrome, a disorder of the connective tissue that affects skeletal, ocular, and cardiovascular systems. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
