Specificity

Clone 2D19 is a ZooMAb® rabbit recombinant monoclonal antibody that detects GPR158. It targets an epitope within 21 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human GPR158.

Application

Quality Control Testing

Evaluated by Western Blotting in human prostate tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GPR158 in human prostate tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected His-tagged recombinant fragment of human GPR158.

Affinity Binding Assay: A representative lot of this antibody bound GPR158 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GPR158 in mouse brain and human gallbladder tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GPR158 in C6 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Metabotropic glycine receptor (UniProt: Q5T848; also known as mGlyR, G-protein coupled receptor 158) is encoded by the GPR158 (also known as KIAA1136) gene (Gene ID: 57512) in human. GPR158 is a member of the G protein-coupled receptor (GPCR) superfamily. It is characterized by seven transmembrane domains, an extracellular N-terminus, and a large intracellular C-terminus. GPR158 is classified as a metabotropic receptor for glycine, although it can also respond to other ligands. Unlike ionotropic receptors, which directly mediate ion flux across membranes, GPR158 influences cellular processes through second messenger systems. It has been implicated in various signaling pathways, including those involved in cell growth and synaptic plasticity. GPR158 is known to interact with G proteins and potentially other signaling molecules, modulating neurotransmitter release and neuronal excitability. GPR158 is highly expressed in the brain, particularly in the cortex and hippocampus. Within neurons, GPR158 is predominantly localized to the post-synaptic density. It is also expressed in other tissues, including the eye and reproductive organs. Dysregulation of GPR158 has been associated with several neurological and psychiatric disorders. Its overexpression has been linked to major depressive disorder, where it is thought to affect the glutamatergic system and synaptic plasticity. In addition, GPR158 have been implicated in the pathophysiology of glaucoma and certain reproductive disorders. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
