Specificity

Clone 1H14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Dual specificity protein phosphatase 6 (DUSP6). It targets an epitope within 24 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal region of human Dual specificity protein phosphatase 6 (DUSP6).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected DUSP6 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected DUSP6 in rat liver tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected DUSP6 in human liver and human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected DUSP6 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound DUSP6 peptide with a KD of 1.3 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Dual specificity protein phosphatase 6(UniProt: Q16828, also known as EC:3.1.3.16, Dual specificity protein phosphatase PYST1, Mitogen-activated protein kinase phosphatase 3, MAP kinase phosphatase 3. MKP-3) is encoded by the DUSP6 (also known as MKP3, PYST1) gene (Gene ID: 1848) in human. DUSP6 is a member of the dual-specificity phosphatases family that serves as an ETS-regulated negative modulator of ERK pathway. It mediates the dephosphorylation of MAPKs on threonine/serine and tyrosine residues. Its tyrosine phosphatase domain is localized in amino acids 206-349. It is essential for the normal dephosphorylation of the long-lasting phosphorylated forms of spinal MAPK1/3 and MAP kinase p38 induced by peripheral surgery, which drives the resolution of acute postoperative allodynia. Dephosphorylation of MAPK1/3 in local wound tissue contributes to resolution of acute pain. It is reported to promote cell differentiation by regulating MAPK1/MAPK3 activity and regulating the expression of AP1 transcription factors. It plays an important role in numerous types of solid tumors as a tumor suppressor and its levels are down-regulated in several types of cancers, including pancreatic, lung, ovarian cancers, and esophageal squamous cell carcinoma. Loss of DUSP6 results in hyperactivation of ERK1/2 that leads to higher cell proliferation, tumorigenicity, and chemoresistance. Two isoforms of DUSP6 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
