Specificity

Clone 1H10 is a ZooMAb® rabbit recombinant monoclonal antibody that detects KLRG1. It targets an epitope within 24 amino acids from the C-terminal, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal, extracellular domain of human KLRG1.

Application

Quality Control Testing

Evaluated by Western Blotting in Ramos cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected KLRG1 in Ramos cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected KLRG1 in lysate from THP-1, Raji cell membrane extract, and human spleen tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1000 dilution from a representative lot detected KLRG1 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected KLRG1 in Ramos cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected KLRG1 in one million Ramos cells.

Affinity Binding Assay: A representative lot of this antibody bound KLRG1 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Killer cell lectin-like receptor subfamily G member 1 (UniProt: Q96E93; also known as C-type lectin domain family 15 member A, ITIM-containing receptor MAFA-L, MAFA-like receptor, Mast cell function-associated antigen) is encoded by the KLRG1 (also known as CLEC15A, MAFA, MAFAL) gene (Gene ID: 10219) in human. KLRG1 is a single pass type II transmembrane glycoprotein characterized by a C-type lectin-like domain. This receptor is encoded by the KLRG1 gene located on chromosome 12 in humans. It features a short cytoplasmic tail that lacks immunoreceptor tyrosine-based inhibitory motifs (ITIMs) but includes a charged residue that may interact with adaptor proteins. KLRG1 functions primarily as an inhibitory receptor on the surface of natural killer (NK) cells and T cells. It is known to inhibit cellular proliferation and cytokine secretion upon ligand binding. The primary ligands for KLRG1 are E-cadherin and N-cadherin, found on epithelial and other cells, which contribute to the immune escape mechanisms of cancer cells. KLRG1 is not universally expressed on all NK or T cells but is found predominantly on the surface of terminally differentiated cells. In the context of T cells, it is mostly expressed on CD8+ effector and effector memory T cells. The expression of KLRG1 is often associated with cellular senescence, particularly in the context of chronic viral infections and cancer. KLRG1 is implicated in various disease states such as chronic infections, autoimmune diseases, and cancers. In cancer, KLRG1′s interaction with E-cadherin can be exploited by tumor cells to evade immune surveillance. In chronic viral infections, high KLRG1 expression is associated with T cell exhaustion, leading to a reduction in the efficacy of the immune response. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
