Specificity

Clone 1H3 is a is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects CXCL9 protein.

Immunogen

GST/His-tagged Human recombinant mature CXCL9 (without the signal peptide).

Application

Quality Control Testing

Evaluated by Western Blotting with GST/His-tagged recombinant human CXCL9.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected recombinant Human CXCL9.

Tested applications

Affinity Binding Assay: A representative lot of this antibody bound recombinant CXCL9/MIG with a KD of 2.3 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CXCL9/MIG in THP-1 cells treated with IFN- and TNF- (both at 100 ng/mL; 24 h).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

C-X-C motif chemokine 9 (UniProt: Q07325; also known as Gamma-interferon-induced monokine, Monokine induced by interferon-gamma, HuMIG, MIG, Small-inducible cytokine B9) is encoded by the CXCL9 (also known as CMK, MIG, SCYB9) gene (Gene ID: 4283) in human. CXCL9 is a small cytokine of the CXC chemokine family that lacks the Glu-Leu-Arg (ELR) domain. It is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form. It is induced in macrophages in response to interferon- and is involved in growth, movement , and activation state of cells that participate in immune and inflammatory responses. It also acts as a chemotactic agent for activated T cells. Its actions are mediated via CXC-chemokine receptor 3 (CXCR3). Overexpression of CXCL9 is reported to reduced tumor progression and metastasis via the inhibition of angiogenesis. CXCL9 is reported to exert its tumor-suppressive function by recruiting tumor-infiltrating lymphocytes in human ovarian cancer. Conversely, CXCL9 can also act directly on tumor cells expressing the CXCR3 receptor to promote cell migration and epithelial mesenchymal transition. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
