Specificity

Clone 2F4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects cytosolic phospholipase A2 (cPLA2). It targets an epitope within 23 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the internal region of human cytosolic Phospholipase A2 (cPLA2).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected cPLA2 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected cPLA2 in lysates from A549 cells and Human testis tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected cPLA2 in Human kidney and 1:100 dilution in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound cPLA2 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Cytosolic phospholipase A2 (UniProt: P47712; also known as cPLA2, Phospholipase A2 group IVA) is encoded by the PLA2G4A (also known as CPLA2, PLA2G4) gene (Gene ID: 5321) in human. cPLA2 is a calcium-dependent phospholipase with lysophospholipase activities. However, it also displays calcium-independent phospholipase A2 and lysophospholipase activities in the presence of phosphoinositides. It preferentially hydrolyzes the ester bond of the fatty acyl group attached at sn-2 position of phospholipids. Its expression is observed in various cells and tissues, including macrophages, neutrophils, fibroblasts, and lung endothelium. It is detected in cytoplasm, nucleus, and Golgi apparatus membranes. It plays a major role in membrane lipid remodeling and biosynthesis of lipid mediators of the inflammatory response. It also plays a role in embryo implantation and parturition through its ability to trigger prostanoid production. It is reported to selectively hydrolyze sn-2 arachidonoyl group from membrane phospholipids, thereby providing the precursor for eicosanoid biosynthesis via the cyclooxygenase pathway. cPLA2 undergoes phosphorylation at serine 505 and serine 727 by MAP kinases in response to mitogenic stimuli. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
