Specificity

Clone 1I15 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CD83. It targets an epitope within 20 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the extracellular domain of human CD83.

Application

Quality Control Testing

Evaluated by Western Blotting with His-tagged, recombinant fragment of human CD83.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant fragment of human CD83.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CD83 in Raji cell lysate.

Affinity Binding Assay Analysis: The binding affinity of this antibody to CD83 peptide was at least one thousand-fold (1,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected CD83 in Raji and Ramos cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD83 in human tonsil tissue sections.

Flow Cytometry Analysis: 1 µg from a representative lot detected CD83 in one million Human peripheral blood mononuclear cells (PBMC).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

CD83 antigen (UniProt: Q01151; also known as hCD83, B-cell activation protein, Cell surface protein HB15) is encoded by the CD83 gene (Gene ID:9308) in human. CD83 is a single-pass type I membrane glycoprotein. It consists of an extracellular V-type Ig-like domain (aa 20-144), a transmembrane domain (aa 145-166), and a cytoplasmic tail (aa 167-205). It is thought to play a role in immune cells activation and immune tolerance. CD83 is expressed on a wide variety of cells, but primarily on the surface of activated immune cells, such as mature dendritic cells, T and B cells. A soluble form of this protein can bind to dendritic cells and inhibit their maturation. Its expression is upregulated upon dendritic cell maturation. Overexpression of CD83 is observed in certain autoimmune diseases while reduced expression or function of CD83 might contribute to immune evasion in cancers and chronic infections. Soluble CD83 has been studied for its potential therapeutic effects, such as prolonging graft survival in transplantation and ameliorating symptoms of autoimmune diseases. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
