Specificity

Clone TROMA-1 is a ZooMAb® rat recombinant monoclonal antibody that specifically detects Cytokeratin-8.

Immunogen

Intermediate filaments extracted from TDM1 Trophoblastoma cells.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Cytokeratin 8 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected recombinant Human Cytokeratin 8.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Cytokeratin 8 in HepG2 cells.

Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated Cytokeratin 8 in HeLa cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound recombinant Human Cytokeratin 8 with a KD of 5.6 x 10-7 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Keratin, type II cytoskeletal 8 (UniProt: P11679; also known as Cytokeratin endo A, Cytokeratin-8, CK-8, Keratin-8, K8, Type-II keratin Kb8) is encoded by the Krt8 (also known as Krt2-8) gene (Gene ID: 16691) in murine species. Cytokeratins belong to a diverse group of intermediate filaments that participate in differentiation and maintain the structural integrity of epithelial cells. Cytokeratins share a common structural organization: a central -helical rod domain flanked by head and tail domains. An important unique feature of cytokeratins is their epithelial cell-type-specific expression. CK8 is a heterotetramer of two type I and two type II keratins. It is expressed in abundance in the epithelia of colon, bladder, ileum, and stomach. CK8 together with KRT19, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle. It is often used together with keratin 18 and keratin 19 to differentiate cells of epithelial origin from hematopoietic cells in tests that enumerate circulating tumor cells in blood. Over-expression of CK8 and K18 is also observed in hepatocellular adenomas and carcinomas and is considered as a reliable marker of preneoplastic lesions arising during mouse hepatocarcinogenesis. CK8 expression has been correlated with malignancy in leukoplakia and carcinomas of the head and neck and its expression has been observed in all non-small-cell lung cancers. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rat monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
