General description

Actin, alpha cardiac muscle 1 (UniProt P68032; also known as Alpha-cardiac actin) is encoded by the ACTC1 (also known as ACTC, ASD5, CMD1R, CMH11, LVNC4) gene (Gene ID 70) in human. Actins are globular multi-functional proteins that serve as the basic building blocks of cytoskeletal microfilaments and are among the most conserved eukaryotic proteins. Six actin types exist, skeletal muscle alpha-actin is encoded by the ACTA1 gene, smooth muscle alpha-actin by the ACTA2 gene, cytoplasmic beta-actin by the ACTB gene, cardiac muscle alpha-actin by the ACTC1 gene, cytoplasmic gamma-actin by the ACTG1 gene, and smooth muscle gamma-actin by the ACTG2 (a.k.a. ACTA3) gene. Although actins show >90% overall sequence homology, isoforms do show spatial, temporal, and tissue-specific expresson patterns and only 50-60% homology is found in their 18 N-terminal residues. Cytoplasmic - and -actins, are thought to be present in all cells, while the other four actin isoforms are typically found in specific adult muscle tissue types. Actins exist in a variety of structural states, depending on the specific ionic conditions or the interaction with ligand proteins. The oligomeric and polymeric forms that actin molecules assume are dependent on the distinct conformations they adopt.

Specificity

Clone 22D3 specifically detected alpha-cardiac actin in rat myocardium homogenates with no cross-reactivity toward - or -smooth muscle actin in rat aorta and chicken gizzard homogenates, cytoplasmic - or -actin in human platelet lysate, nor -skeletal actin in rat skeletal muscle homogenate (Driesen, R.B., et al. (2009). J. Cell. Mol. Med. 13(5):896-908).

Immunogen

KLH-conjugated linear peptide corresponding to human mature alpha-cardiac actin N-terminal end sequence with an acetylated N-terminus.

Application

Immunocytochemistry Analysis: A representative lot immunostained 4% paraformaldehyde-fixed, 0.05% NP-40-permeabilzed rectangular rabbit cardiomyocytes by indirect immuoflourescence (Driesen, R.B., et al. (2009). J. Cell. Mol. Med. 13(5):896-908).

Immunofluorescence Analysis: A representative lot immunostained cardiomyocytes in human left ventricular endocardial biopsies, rabbit left ventricular myocardium, as well as in goat atrial myocardium frozen tissue sections by fluorescent immunohistochemistry (Driesen, R.B., et al. (2009). J. Cell. Mol. Med. 13(5):896-908).

Western Blotting Analysis: A representative lot specifically detected alpha-cardiac actin in rat myocardium homogenate with no cross-reactivity toward - or -smooth muscle actin in rat aorta and chicken gizzard homogenates, cytoplasmic - or -actin in human platelet lysate, nor -skeletal actin in rat skeletal muscle homogenate (Driesen, R.B., et al. (2009). J. Cell. Mol. Med. 13(5):896-908).
