General description

Nitric oxide (NO) is a highly reactive radical that plays a crucial role in numerous essential physiological functions. It is produced by the oxidation of arginine through nitric oxide synthase (NOS), and it actively participates in various processes, including host defense, activation of regulatory proteins, and direct covalent interactions with functional biomolecules. Inhibition of NOS can have diverse biological effects, particularly within the cardiovascular system. Simple, direct and non-radioactive procedures for measuring NOS are becoming popular in drug discovery, biomedical and biochemical research.

The mechanism of action for the Nitric Oxide Synthase Inhibitor Screening Kit comprises two steps:

  • NOS Reaction Step: In this step, nitric oxide (NO) is produced through the activity of nitric oxide synthase (NOS).
  • NO Detection Step: Following NO generation, the produced NO undergoes rapid oxidation to form nitrite and nitrate. The measurement of NO production is then conducted by quantifying the reduction of nitrate to nitrite, employing an enhanced Griess method.

Principle
