Biochem/physiol Actions

Sulopenem is an orally available penem antibiotic designed to treat Gram-negative multi-drug resistant infections. It can be administered as an intravenous preparation, and it is also orally available.
Sulopenem, formerly CP-70,429, is stable to renal dehydropeptidase I and hydrolytic activity of AmpC β-lactamases and extended-spectrum β-lactamases (ESBLs) that convey resistance to third-generation cephalosporins. This action of sulopenem helps to act against urgent drug-resistant antimicrobial threats, including ESBL-producing Enterobacteriaceae. Sulopenem is effective than ciprofloxacin for treating urogenital upper urinary tract infection (uUTI) in adult women due to quinolone non-susceptible pathogens.

The in vitro activity of sulopenem was assessed against a collection from 2014 to 2016 of 539 urinary isolates of Escherichia coli from Canadian patients by using CLSI-defined broth microdilution methodology. A concentration of sulopenem 0.03 µg/ml inhibited both 50% (MIC50)

In order to deal with the rising problem of antibiotic resistance, newer antibacterials are being discovered and added to existing pool. Since the year 2000, however, only four new classes of antibacterials have been discovered. These include the oxazolidinones, glycolipopeptides

For 297 penicillin-susceptible, -intermediate, and -resistant pneumococcal strains, the sulopenem MIC(50)s were 0.008, 0.06, and 0.25, respectively, and the sulopenem MIC(90)s were 0.016, 0.25, and 0.5 microg/ml, respectively. The MIC(50)s of amoxicillin for the corresponding strains were 0.03, 0.25, and
