Application

NVP-ADW742 has been used as an insulin-like growth factor 1 receptor (IGF1R) inhibitor to test the effects of filamin A (FLNA) silencing in a cell proliferation assay.

Biochem/physiol Actions

NVP-ADW742 is a potent and selective ATP-competitive inhibitor of Insulin-like growth factor-I receptor (IGF-IR) kinase. NVP-ADW742 sensitizes cancer cell lines to multiple anticancer agents including etoposide, carboplatin, imatinib, vincristine, and doxorubicin.

Adrenocortical carcinomas (ACCs) overexpress insulin-like growth factor 2 (IGF2), that drives a proliferative autocrine loop by binding to IGF1R and IR, but IGF1R/IR-targeted therapies failed in ACC patients. The cytoskeleton actin-binding protein filamin A (FLNA) impairs IR signalling in melanoma
