General description

Specificity

Clone 2J22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects human Monocarboxylate transporter 4 (SLC16A3). It targets an epitope within 22 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal, cytoplasmic domain of human Monocarboxylate transporter 4 (SLC16A3).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SLC16A3/MCT4 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SLC16A3/MCT4 in A431 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound SLC16A3/MCT4 peptide with a KD of 3.5 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SLC16A3/MCT4 in A431 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SLC16A3/MCT4 in human skeletal muscle tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Monocarboxylate transporter 4 (UniProt: O15427; also known as MCT 4, Solute carrier family 16 member 3) is encoded by the SLC16A3 (also known as MCT3, MCT4) gene (Gene ID: 9123) in human. MCT4 is a multi-pass membrane protein with seven cytoplasmic domains, twelve transmembrane domains, and six extracellular domains with amino and carboxyl termini facing the cytosol. It contains two basolateral sorting signals (aa 423-441; 441-465) in its C-terminal cytoplasmic tail that are essential to direct MCT4 to the basolateral membrane. It is the major monocarboxylate transporter present in skeletal muscle that mediates the transport of lactate, pyruvate, and ketone bodies across the plasma membrane. It is also detected in tumor cells and white blood cells that have high glycolytic rate. It displays higher transport efficiency for L-lactate when compared to D-lactate and pyruvate (Km = 28, 513, and 153 mM, respectively in Xenopus oocytes). Human MCT contains six conserved arginine residues and arginine 278 is reported to be the crucial residue involved in L-lactate recognition. Any mutation in this residue is shown to void L-lactate transport. High levels of MCT4 are reported in head and neck squamous cell carcinoma (HNSCC) and these higher levels are associated with poor prognosis. It is not expressed in normal oral mucosa but is detected in the transformed epithelium. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
