Specificity

Clone 3H4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects E3 ubiquitin-protein ligase XIAP. It targets an epitope within 24 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal region of human XIAP.

Application

Quality Control Testing

Evaluated by Western Blotting in COS-7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected XIAP in COS-7 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected XIAP in C6 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected XIAP in A431 and HepG2 cells..

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected XIAP in human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound XIAP peptide with a KD of 5.0 x 10-8 in an affinity binding assay.

Target description

E3 ubiquitin-protein ligase XIAP (UniProt: P98170; also known as EC:2.3.2.27, Baculoviral IAP repeat-containing protein 4, IAP-like protein, ILP, hILP, Inhibitor of apoptosis protein 3, IAP-3, hIAP-3, hIAP3, RING-type E3 ubiquitin transferase XIAP, X-linked inhibitor of apoptosis protein, X-linked IAP) is encoded by the XIAP (also known as API3, BIRC4, IAP3) gene (Gene ID: 331) in human. XIAP is one of the most potent anti-apoptotic IAP family member that contains three BIR domains (aa 26-93; 163-230; and 265-330). It is reported to directly neutralize caspase-9 via its BIR3 domain and the effector caspases-3 and -7 via its BIR2 domain. The BIR3 domain interacts with the Apaf-1/caspase-9 holoenzyme by sequestering the N-terminus of the small subunit of processed caspase-9. In addition to its role in regulating caspases and apoptosis, it also modulates inflammatory signaling and immunity, mitogenic kinase signaling, cell proliferation, and cell invasion and metastasis. It can directly bind to the active site pocket of caspase 3 and 7 and obstructs substrate entry. Aberrant expression of XIAP is reported in a variety of human cancers and it mediates cellular resistance to chemotherapeutic drugs. XIAP also acts as an E3 ubiquitin-protein ligase and regulates NF- B signaling. Other target proteins for its E3 ubiquitin-protein ligase activity include: RIPK1, MEKK2, DIABLO/SMAC, and Survivin. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
