General description
Enolase enzyme contains three subunits- α, β, and γ-and five isozymes namely αα, ββ γγ, αγ, and βγ. The isozymes that contain a γ subunit are expressed in neuronal and endocrine tissue and are known NSEs (neuron-specific enolases). NSE or γ-enolase is a highly soluble intracellular enzyme, which resides in cytoplasm, and shows predominant expression in neuronal tissues.
Neuron-specific enolase (NSE) is a glycolytic isoenzyme which is located in central and peripheral neurons and neuroendocrine cells. This enzyme is released into the CSF when neural tissue is injured. Neoplasms derived from neural or neuroendocrine tissue may release NSE into the blood. NSE is a useful substance that has been detected in patients with certain tumors, namely - neuroblastoma, small cell lung cancer, medullary thyroid cancer, carcinoid tumors, pancreatic endocrine tumors, and melanoma. NSE shows neurotrophic and neuroprotective properties on a broad spectrum of central nervous system (CNS) neurons and binds in a calcium-dependent manner to cultured neocortical neurons promoting cell survival.
Biochem/physiol Actions
Enolases are responsible for catalyzing the catabolism of glycerol in the glycolytic pathway, and NSE catalyzes the conversion of 2-phosphoglycerate into phosphoenolpyruvate. NSE (neuron-specific enolase) is involved in the tumorigenesis of neuroendocrine origin tumors. Patients with multiple myeloma (MM) show elevated NSE serum levels, and these levels might help prognosticate efficiency of chemotherapy and disease progression. In diabetic patients the serum levels of NSE are higher than in normal subjects, and these levels are linked with peripheral neuropathy in diabetic patients. The serum levels of this protein are also elevated in patients with traumatic brain injury and neurodegenerative disease. In young adults, reduced hippocampal GM (gray matter) is related to increased NSE levels in obese individuals, suggesting an association between obesity with a specific marker of neuronal injury in a neurodegeneration and cognitive decline-sensitive brain area.
