Specificity

Clone 2F2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Tubulin. It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Tubulin.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Tubulin in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Tubulin in lysate from A431 cells and 1:1,000 dilution in lysates from C6, NIH3T3, and COS-7 cells..

Affinity Binding Assay: A representative lot of this antibody bound Tubullin peptide with a KD of 1.8 x 10-7 in an affinity binding assay.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected Tubulin in one million Hela and A431 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Tubulin in A431, HeLa, HUVEC, and NIH3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Tubulin in Human testis tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Tubulin alpha-3E chain (UniProt: Q6PEY2; also known as Alpha-tubulin 3E) is encoded by the TUBA3E gene (Gene ID: 112714) in human. Tubulin is the major constituent of microtubules (MTs). MTs are composed of α/β-tubulin heterodimers that self-assemble head-to-tail to form protofilaments and laterally to form a hollow tube. α/β heterodimers associate head-to-tail to form protofilaments running lengthwise along the microtubule wall with the β-tubulin subunit facing the microtubule plus end conferring a structural polarity. Tubulin binds two moles of GTP, one at an exchangeable site on the β-chain and one at a non-exchangeable site on the α-chain. Tubulin dimers constantly polymerize and depolymerize, and MTs can undergo rapid cycles of assembly and disassembly. MTs also undergo treadmilling, in which tubulin molecules bound to GDP are continually lost from the minus end and are replaced by the addition of GTP-bound tubulin molecules to the plus end of the same MT. Various post-translational modifications, including acetylation, polyglutamylation, polyglycylation, detyrosination, phosphorylation, and palmitoylation, can regulate the polymerization properties of tubulins and/or their interactions with microtubule associated proteins and motor proteins. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
