Application

5-DBCO-PEG4-dUTP is recommended for two-step labeling of DNA/cDNA e.g. by PCR with Taq polymerase. It is enzymatically incorporated into DNA/cDNA as a substitute for its natural counterpart dTTP. The resulting DBCO-functionalized DNA/cDNA can be labeled with azide-labeled molecules using copper-free click chemistry. This enables experiments such as introduction of a biotin group for purification tasks, introduction of a fluorescent group for detection, or crosslinking the DNA to azide-functionalized biomolecules such as proteins.
