General description

EMD Millipore’s AOF ITS (100X) supplement is an animal-origin-free product that contains human recombinant insulin, human recombinant transferrin along with sodium selenite.

Undefined animal derived media supplements, such as fetal bovine serum, have recently been an area of concern for many cell culturists due to their increased costs, lot to lot variability and increased regulation worldwide. The elimination of fetal bovine serum from cell culture media provides a more defined enviroment to study basic biological questions. Serum-free and reduced serum formulations often require purified forms of the factors normally supplied by serum that are suitable for in vitro growth and differentiation studies such as insulin, transferrin and selenite.

ITS supplements have been proven to replace serum and grow the following cells in serum free or reduced serum conditions: CHO, HEK293, fibroblasts, hybridoma, VERO cells, Hela, HT29, MDCK, Mesenchymal, IPS Cells, Cardiac, kidney, keratinocyte, embryonic, neural stem cell, hepatocytes.

Components

All components are derived using AOF (Animal-Origin-Free) reagents and protocols.

Recombinant Human Insulin is derived from Pichia pastoris (yeast).
Recombinant Human Transferrin is derived from Oryza sativa (rice).
Sodium Selenite is a synthetic chemical.

Quality

Tested for the ability to support the proliferation of VERO cells in serum free conditions over a 3 day time period.
pH: 7.6-8.0.
Product is sterile.
Endotoxin levels are ≤ 1 EU/mg.

Physical form

Product is made in an Earle′s Balanced Salt Solution and filtered through a 0.22 micron membrane before vialing. Solution is a light pink clear solution that is free of particulates.

Storage and Stability

Store at 2-8°C. Further dilutions should be made into appropriate buffer and/or medium to which protein has been added, such as DMEM/1% Human Serum Albumin or equivalent.
